氧化应激状态评价的生化指标

生物体进行呼吸代谢等氧化反应会产生活性自由基,在正常机体环境下,自由基具有稳定的清除系统,使得机体自由基含量保持较低浓度的平衡。氧化应激(Oxidative Stress, OS)是指机体由于受到内源性和(或)外源性刺激,使得体内氧化与抗氧化作用间的平衡失常,自由基产生过多。过多的自由基在体内产生一系列负面作用,可氧化损伤生物分子,并进一步引起细胞死亡和组织损伤等,与很多病理过程相关。

很多疾病均与氧化应激相关,但还不十分清楚氧化应激是疾病的原因还是疾病过程的一种结果。因自由基寿命短暂,且缺乏足够敏感的技术直接检测生物体内的自由基,因此需建立一套衡量体内氧化应激状态的评价体系,为防治氧化损伤提供支持。本公司针对评价氧化应激状态的一系列问题,精心选择与之密切相关的检测指标,建立出一套较为完整的评价体系,并逐一攻克研发出与之对应可行的检测方法,以供科研用户选择。

2、非酶抗氧化剂

非酶抗氧化剂主要是可清除自由基的低分子量物质,包括谷胱甘肽、抗氧化维生素、泛醌还原物、金属硫蛋白及硫氧还蛋白等。

谷胱甘肽(GSH)是一种由谷氨酸、半胱氨酸和甘氨酸结合的三肽,GSH的结构中含有一个活泼的巯基(-SH),易被氧化脱氢,这一特异结构使其成为体内主要的抗氧化剂。而且GSH是多种酶如GSH-Px的辅酶,涉及多种生物学过程,参与清除ROS,防护氧化应激对机体产生的损伤。GSH的含量可在一定程度上反映机体的抗氧化能力。

抗氧化维生素包括Vit C、Vit A、Vit E及β-胡萝卜素,其中以Vit E和Vit C为重要。Vit E是一种天然脂溶性抗氧化剂,能阻断机体不饱和脂肪酸的过氧化,维持不饱和脂肪酸细胞膜的完整性和正常功能,并可清除超氧阴离子自由基。Vit C(又称抗坏血酸,ASA)作为植物细胞中重要的抗氧化剂,ASA在保护叶绿体免于氧化损伤起着举足轻重的作用,也是衡量农作物产品品质的重要指标之一。

其他抗氧化剂还包括尿酸、铜蓝蛋白(Cp)等。尿酸为嘌呤代谢的终末产物,尿酸水平的异常会导致多种疾病如痛风、肾病、心血管疾病的发生;此外,尿酸还是重要的抗氧化剂,能清除超氧化物,羟自由基等。铜蓝蛋白是血浆的含铜蛋白,有运输铜的功能,同时具有氧化酶的活性,也是细胞外液重要的抗氧化剂。

3、机体的抗氧化能力

单独测定某一种或某些抗氧化成分的含量往往不能全面的反映组织的抗氧化能力,且在操作上较为困难与或繁琐,因而可通过T-AOC来评价机体总体上的抗氧化能力,即测定对象中各种抗氧化物质和抗氧化酶等构成总抗氧化水平。

羟自由基是人体在新陈代谢过程中产生的对生物体毒性强、危害大的一种自由基。它可以使组织中的糖类、氨基酸、蛋白质、核酸等物质发生氧化,遭受氧化性损伤和破坏,导致细胞坏死或突变。羟自由基清除能力是样品抗氧化能力的重要指标之一,在抗氧化类保健品和药品研究中得到广泛应用。
